  • Two methods (Genomic DNA extraction kit and Phe/Chl)were used to extract genomic DNA from fresh muscles, livers and caudal fins, and alcohol-preserved ones of bighead carps. The results showed that both of the methods can extract high-quality DNA from the three organizations under different conditions. The genomic DNA can be used for the further molecular biology research. The DNA samples of caudal fins preserved in alcohol are the best quality, followed by muscle, liver the worst. The results showed the DNA extraction kit could obtain high purity genomic DNA with low density, and high concentration, good quality and relatively low cost by Phe/Chl method. The phenol extraction of genomic DNA from alcohol-preserved caudal fin is an effective method of promoting and using in molecular studies on fish.
