鳙鱼不同组织基因组DNA提取方法的探讨

Comparison of genomic DNA extraction of Aristichthys nobilis

  • 摘要: 分别采用试剂盒和常规苯酚/氯仿抽提法提取新鲜的鳙鱼肌肉、肝脏、尾鳍以及用乙醇保存的3种组织基因组DNA,并对其进行综合分析。结果表明,对3种组织的新鲜样品2种方法均能提取高质量的DNA,而用乙醇保存的3种组织,DNA样品质量最好的是尾鳍,其次是肌肉,肝脏最差;总的来看,试剂盒法提取的DNA纯度较好,但是浓度低,而苯酚法得到的DNA浓度高,质量好,且相对成本低。综合比较,苯酚/氯仿抽提法提取乙醇保存的尾鳍组织基因组DNA是一种行之有效的方法,可以在鱼类分子研究中推广应用。

     

    Abstract: Two methods (Genomic DNA extraction kit and Phe/Chl)were used to extract genomic DNA from fresh muscles, livers and caudal fins, and alcohol-preserved ones of bighead carps. The results showed that both of the methods can extract high-quality DNA from the three organizations under different conditions. The genomic DNA can be used for the further molecular biology research. The DNA samples of caudal fins preserved in alcohol are the best quality, followed by muscle, liver the worst. The results showed the DNA extraction kit could obtain high purity genomic DNA with low density, and high concentration, good quality and relatively low cost by Phe/Chl method. The phenol extraction of genomic DNA from alcohol-preserved caudal fin is an effective method of promoting and using in molecular studies on fish.
