液相色谱串联电喷雾质谱检测中溶剂对硝基呋喃类代谢物响应的影响

Influence of solvent on nitrofuran metabolites response by liquid chromatography-tandem mass spectrometry

  • 摘要: 通过对比不同溶剂对液相色谱串联电喷雾质谱检测硝基呋喃类代谢物灵敏度的影响, 以筛选最佳的流动相和上机溶液。Q1扫描时, AHD在负离子模式下有最高的响应强度。正离子模式检测时, 甲醇中添加0.1%的氨水, 4种化合物均能获得令人满意的灵敏度。用液相色谱串联正电离电喷雾质谱检测时, 以甲醇和0.1%的氨水作为流动相, 上机溶液中也添加0.1%的氨水, 灵敏度高, 杂质干扰少, 准确度好, 能够较好地满足鱼、虾、蟹等复杂样品基质中痕量硝基呋喃类代谢物残留检测的需要。

     

    Abstract: To select the optimummobile phase and injection, the influence of solvent on sensitivity of nitrofuran metabolites (AMOZ,SEM, AHD and AOZ) was investigated by liquid chromatography tandem mass spectrometry (LC-MS/MS). The highest intensity of AHD was obtained in Q1 scan with negative mode. However, the satisfactory sensitivities of four nitrofuran metabolites could be gained in positive mode while the compounds were dissolved in methanol that contained 0.1% aqueous ammonia. Gradient elution for LC separation was performed by using methanol and 0.1% aqueous ammonia in water solution, and the injection was also added with 0.1% aqueous ammonia. The optimized method is suitable for determining of four nitrofuran metabolites in multiple matrices with high sensitivity, less interference and excellent accuracy.
