高效液相色谱法测定水产品中孔雀石绿残留量的的优化研究

Optimization of HPLC determination of malachite green residue in aquatic products

  • 摘要: 采用高效液相色谱法测定水产品中孔雀石绿的残留量,优化了检测过程中的提取方式、硼氢化钾用量、蒸馏温度、固相萃取柱选择和流动相选择等对加标回收率的影响因素。样品经匀浆与超声相结合提取,用质量分数为6%的硼氢化钾还原,40 ℃真空蒸干,PRS固相萃取柱净化,所得样液以V(乙腈):V(乙酸铵)=75∶25的缓冲液为流动相进行测定。结果表明,样品回收率高,且前处理简便,结果准确。

     

    Abstract: We used high performance liquid chromatography to determine malachite green (MG) in aquatic products, and optimized the factorsaffecting the recovery, such as extraction method, KBH4 amount, evaporation temperature, selection of solid phase extraction column and mobile phase, etc.. The sample was extracted by homogenate and ultrasonic instrument, reduced with 6% of KBH4, vacuum-evaporated at 40 ℃, purified with PRS solid phase, and determined by the mobile phase of 75% acetonitrile with 25% ammonium. Thus, weobtain an accurate method which has simple pretreatment but high recovery.
