赤点石斑鱼诺达病毒的纯化及其衣壳蛋白的Western-blot分析

Purification and Western-blot assay of the capsid protein of the red-spotted grouper nodavirus

  • 摘要: 采用差速离心,蔗糖(10%~40%,W/V)密度梯度离心和氯化铯(30%~40%,W/W)等密度梯度离心法,对赤点石斑鱼诺达病毒大亚湾株(RG-CN)进行了纯化,测定计算其浮密度为1.3102~1.3243 g·cm-3。通过Western-blot法检测到的病毒的结构蛋白的分子量为37和31 kDa,而以31 kDa的蛋白为主。

     

    Abstract: Red-spotted grouper nodavirus Dayawan isolate(RG-CN)was purifed by differential centrifugation, sucrose density gradient centrifugation(10%~40%, W/V) and CsCl isopyknical density centrifugation(30%~40%, W/W).The results showed that the viral particles exhibited a buoyant density of 1.3102~1.3243 g·cm-3. By Western-blot assay, viral capsid protein showed two polypeptides with molecular weights of 37 and 31 kDa, mainly being 31 kDa.
