微生物法检测水产品中粘杆菌素的残留

Microbiological inhibition method for determination of colistin residues in muscles of aquatic products

  • 摘要: 以支气管炎鲍特氏菌(Bordetella bronchiseptica)为检测菌种, 样品经甲醇-0.1%甲酸水(体积比3:7)超声提取, 固相萃取柱净化, 氮吹浓缩, 用杯碟法测定水产品草鱼(Ctenopharyngodon idellus)、斑节对虾(Penaeus monodon)、中华鳖(Pelodiscus sinensis)、日本鳗鲡(Anguilla japonica)和蟹(Brachyura)肌肉中粘杆菌素的残留。结果显示, 粘杆菌素在各水产品肌肉组织中0.4~2.4 μg·mL-1范围内药物浓度对数与抑菌圈直径的线性关系良好, 相关系数R2>0.990 0;在0.20 μg·g-1、0.40 μg·g-1和0.60 μg·g-1的添加水平下, 粘杆菌素的平均回收率为70.59%~93.39%, 变异系数为3.22%~14.01%。测定低限草鱼和对虾肌肉为100 μg·kg-1, 鳖和鳗鲡肌肉为75 μg·kg-1, 蟹肌肉为125 μg·kg-1。研究结果表明, 运用微生物法检测水产品中粘杆菌素的残留是可行的, 方法最低检测限达到农业部颁布的残留限量标准。

     

    Abstract: Bordetella bronchiseptica were extracted with methanol-0.1%formic acid water (volume ratio 3:7) under ultrasonication, purified by solid phase extraction column, and blowed with nitrogen gas. Then we used B.bronchiseptica to detect the residues after the concentration of samples. Results show that colistin in the muscle tissue of aquatic products ranged from 0.4 μg·mL-1 to 2.4 μg·mL-1, with good linearity between drug concentration logarithm and the diameter of bacteriostatic (R2> 0.990 0). Colistin in muscle was fortified at 0.20 μg·g-1, 0.40 μg·g-1 and 0.60 μg·g-1, and the average recovery was 70.59%~70.59%, while the coefficient of variation was 3.22%~14.01%. The limits of determination were 100 μg·kg-1 in grass carp and prawn muscles, 75 μg·kg-1 in turtle and eel muscles, and 125 μg·kg-1 in crab muscle. The cylinder plate method for determination of colistin residues is good. All minimum detection of drugs meet the national standard of residue limits issued by Ministry of Agriculture of China.
